Most ingredients marketed for relaxation ask for a trade-off: less alertness in exchange for less tension. L-theanine's research base points to something different — a calming effect without the accompanying fog. It's also one of the more thoroughly tested compounds in this category, with human studies measuring brain activity and stress hormones directly, rather than relying on self-reported mood alone.
What it does
L-theanine crosses into the brain and is associated with an increase in alpha brain wave activity — the pattern typically seen in a relaxed, alert state, similar to what's measured during meditation (Human Clinical Studies database). Researchers describe this as "wakeful relaxation": calm without drowsiness.
An early EEG study found this effect was strongest in participants who scored higher on anxiety measures beforehand — suggesting the response is proportional to existing tension, rather than a uniform sedative effect (Kobayashi et al., cited in Human Clinical Studies).
The evidence on stress hormones
A 2021 study measured something more concrete than mood: cortisol, the body's primary stress hormone. Healthy adults completed a validated stress task, then received either L-theanine or a placebo. The L-theanine group showed a significantly greater increase in relaxed brain wave activity, alongside a measurable drop in cortisol one hour later (published in *Nutrients*, 2021). That combination — a physiological stress marker responding, not just a subjective sense of calm — is a stronger form of evidence than most ingredients in this category can point to.
What regular, lower-dose use looks like
Single-dose lab studies are designed to detect a strong signal under controlled stress; daily use is a different question. A 56-day study using just 50mg of L-theanine per day still found a significant reduction in stress, measured on a validated scale (cited in a 2024 PMC review) — evidence that meaningful effects don't require the larger doses used in single-session testing.
A separate 28-day study, using a higher 200mg daily dose, found a reduction in light sleep with no negative effect on next-day cognitive performance, suggesting sleep quality was maintained rather than disrupted (PMC, 2024).

Where the research is still developing
A 2025 review noted that while these effects are consistently reported, results across studies vary, and called for larger, more rigorous trials ([PMC, 2025]()). L-theanine is well-supported, not fully settled — an honest distinction worth holding onto.
